I am replacing my 7 year old dell windows 2000 computer (with NOD32 installed) with a new vista home premium computer.

I want to be protected while I am making the switchover(another couple of weeks) but I don't want to purchase an additional subscription.

I am thinking I would get the NOD32 30 day trial version then after the migration is complete uninstall NOD32 from my old computer and the activate my subscription on the trial version.

Would I be able to use both of them at the same time if one was only the trial version?

yes, and you can uninstall NOD32 licensed version of the old computer later,
and use the AV- etc. license info to download te version for Vista and replace the trial there with your current license.
According to the license of NOD32 you can run it on 1 computer.
It doesn't matter if it is on the old of new one, just don't run it on 2.
