When I click NOD32 under Threat Protection Modules I am given the following scan options: Local, Run NOD32, In-Depth Analysis and Diskettes.

    Local: Scans local disks
Run NOD32: Runs an on demand scanner
In-depth analysis: Performs an in-depth analysis
    Diskettes: Scan inserted diskettes

Do all the above play a different role?

Also with the HDD scanners whats the best one to use, say if I want to do a quick scan for my drives being C and my external USB drive.

I've heard that NOD32 has got very fast scanning rate but how do I use this function to activate the fast scanning rate. Say for eg. from the above choices whats the best one to use for a fast scan.

    Thank you.

    Yes, they are different...

Local will limit it's scan to local hard drives - no network drives scanned
In-Depth with run with most options enabled, both local and network I think - see the "In Depth" profile for more information.
Scan Diskettes is like setting up NOD32 as a cleaning station
Run NOD32 will start NOD32 with the control center profile
