When I connect a 3TB drive INTERNALLY and set it up as GPT, will I be able to read/write it externally (via USB)?

Yes, if the disk is initialized using Windows 7 x64. No need to use Paragon GPT Loader.

And if I initialized it internally with WinXP+ParagonGPTLoader, will I be able to use it externally (USB) with WinXP or with Win7?

GPT Loader doesn't work with USB devices.
It works with single 3TB drives connected directly via SATA to motherboard.
