I have setup several ERAS servers with no issue, finally decided to get one myself for home use. Can't seem to get the update mirror working.

    Setup:
Windows XP SP3 Pro VM on ESXI 4.1 server, no firewall on, member of local domain (homenet.local)

Updates -> Update Server: Choose Auto
    Username/PW Set
Able to get updates from ESET site
    Create update mirror checked
    Directory set to C:\NOD32 Mirror\
    Provide update files .... checked
    HTTP Port - 2221
    Auth - None

If I try to browse to http:\\servername:2221 I get ISE 500
If I try to browse there on the WinXP itself using http:\\localhost:2221 I get the same error

There are no other web servers running on this box, and no internal firewalls online.

    Any Suggestions?

Bingo sir, stupid Hosts file was hosed ... you made me think of that. I just restructured my network so the IP's in Hosts on my Mac was a bit hosed.. the local was the a different issue, but it was a messup on my part.
