Can I enable protection of the entire C drive against any writes without having any malfunction of my PC?
And is it realistic to enable protection of the entire C drive against any writes?

considering that DS is mainly supposed to be a BB, id say keep it on the sensitive areas that it monitors by default otherwise your basically making it like any other HIPS.

You may as well use something like returnil if you want to protect C: against permanent writes.
