I was a little bit irritated with this. Everytime I open up and log in in my laptop my NOD32 antivirus automatically make virus updates even if I haven't connected it yet to the internet and even if I have already configured and unchecked all the automatic update in the scheduler setting.

Is there anything that I can do so that it will not automatically updates everytime I log-in in my laptop?

I'd strongly recommend posting ESET-related questions in our official forum at https://forum.eset.com where they will receive better attention from ESET's moderators and advanced users.

As for automatic updates, with v10 installed updates should be run automatically only if a network connection is detected. Make sure that you have the latest v10.1 installed. The default regular update task in Scheduler should stay enabled at all times in order for the ESET product to be constantly updated and providing maximum protection against newly emerging threats.
