    I have noticed that ESS 5 slows down VirtualBox a lot, and sometimes causes 100% CPU activity. What exclusions can I set to have it just ignore VirtualBox?

    A good start would be excluding both the Virtualbox directory, and the virtual machine directories.

    Does disabling real-time protecton, firewall or self-defense make a difference?

    It does. If I disable real time protection and the firewall it runs MUCH faster and the CPU stays below 10% most of the time. This happens on both of my PCs.
