I am using the 30 Trial version of NOD32 Version 3 and I have been reading the various multi license threads. If I purchase a single user license but install NOD32 Version 3 on multiple PC's how does ESET know that I am using the single license version on more than one PC?

because your updates are grabbed from the update servers by each computer - when it is seen that multiple machines are using the same license number, it's easy to shut that license off - then you're required to contact customre support to ask what's wrong, and they tell you that multiple machines were using a single machine license.

The 3 machine license is relatively cheap anyway....
