HELP!

I am having a problem geting rid of a site that has highjacked my computer. It has taken over my desktop and everytime i open internet explorer it changes my home page to it's home page.....some dinamo direct site.
can anyone help me get rid of this.....have run antivirus and spyware but have had no luck
