If I start DD10 from within Vista, and then set up any operations that require a reboot, the system fails to do them. Rebooting the system simply starts Vista again. Could this be a BIOS setting issue? It's a new Asus P6T motherboard.

The Full Mode (Linux-based) version of DD on the CD most likely doesn't include the necessary drivers for your motherboard/chipset. This version can also be used (depending on the procedures being performed) when a reboot is required from Windows. If DD can't find any drives or detects any errors, etc., it will abort and no changes will be made.
If you want the Full Mode version to function and the Windows-reboot version to function, you'll need to contact Acronis Support and see if they can provide you with a custom build that includes the drivers. Note that it is generally recommended by users to do any procedures that would require a reboot by booting to the DD CD instead of from Windows. This gets Windows out of the procedure and also lets you have more control over the steps since you can see what's being done.
