I had the same problem with an earlier version of 4 with Vista (on 2 pc's). I have to run V3.
Try booting into the Safe Mode once, then restart with a normal boot.
I would not discount the fact that you may have malware preventing NOD from starting. You may want to try a good checkup and cleaning if...

If you search, others have reported the exact same symptom you describe. Remember, you can always use version 3 until v4 matures a little more.
