Disk De-Frag with/without Protection?

Should a disk de-fragmentation be run with RETURNIL protection off or can it be run with it on? My de-frag program is set to run in auto mode while the system is idle. Would these modifications be lost after a reboot?

You should not try to defragment when Returnil protection is on. Please take some time and read through the existing threads as this and a great deal more useful information is available.
