I am trying to do a backup of the system partition using the one-click protection but keep getting an error stating:

"The destination you have chosen does not have enough space for a system backup archive. The default location will be used."

The system partition C: drive is a 145GB partition with 55GB of used space. The external USB drive is a empty 300GB drive. Why am I getting this error?

According to Acronis User Guide, what you're describing is not supposed to happen. If you haven't already, contact TechSupport and create a ticket to ensure that Aronis knows about this. Meanwhile, someone here might be able to give you a work around better than mine, which is to create task to make a full backup of your desired source to your desired target -- it's not one click and not what you paid for but will probably work until Acronis can fix.
